Essentially, I have Night Light enabled in the panel to change my colour temperature
Occasionally (every few minutes) I get a brief flicker or two where the temperature switches back to what it was when Night Light was off, and then back again.
Why is this conflict happening?
What could be fighting in the display settings that I have overlooked?

The screen of my laptop (Dell XPS 15) is rather blue.
An alternative could be to set the preinstalled D50 color profile to the laptop screen on the standard Settings app's Color sidebar item. (Monitor calibration misery is not needed; the D50 profile is available from the Add profile dialog).
This in itself compensates the blue-ness of the LCD to a really decent level.
It is not timed or periodic, it's on all the time. This could be your "day-time" compensation.
The night-light, when it gets turned on, will add its shift on top of this; they play nicely together.
If, for whatever reason, Flux, Redshift or the Night Color applet do not work for you, consider installing sct from the universe repository provided you're on 18.04 or higher. sct isn't available for 16.04 or older versions.
It's a small program and probably won't pull in many dependencies. You can try apt install -s sct to preview what will be installed: -s provides a simulation and doesn't need sudo.
If you do decide to try sct, man sct has

sct sets the screen's color temperature in a range from 1000 to 10000
OPTIONS
temperature
If passed a value in the correct range (see above) sct will set the current screen temperature to this value
-h If -h is passed sct will display usage information
none If no options are passed sct sets the color temperature to the default of 6500
So when you want a warmer screen, open a terminal and run sct 3500 for example. And when you want to revert to normalcy, just run sct.

I've had that same problem and was able to find the source of the problem.
I had another program installed before 18.04 called "Red Shift" that was conflicting with the "Night light" feature. I removed that program and the issue was fixed since then.
You should check if you do not have another program that tries to manage the screen at the same time.
