I have a lot of images on my laptop as I work in graphics. On the same system, I also have a folder containing my family pictures.
I would like to do two things:

This blog post by Andrea Righi suggests that there isn't a config option in the kernel, but it also sketches out a little glibc wrapper library to just set the O_DIRECT flag on every call to open().
This should have the same effect as disabling the read-buffer completely.
Additionally, the OpenOffice wiki has a couple of methods to do cold-start testing. This isn't what you want, but I thought I include it anyway.
Thumbnail cache is limited to 512MB, so several hundred megabytes sounds logical. It also shouldn't keep files older then 180 days.
If you want to change it, try gconf-editor, /desktop/gnome/thumbnail_cache.
Exactly the same solution is aplicable to 12.04. You may need to install gconf-editor, other then that, nothing has changed.
Update
For 14.04 you need to install dconf-tools, and launch dconf-editor.

Thumbnails are stored in
~/.cache/thumbnails. Simply deleting everything in there will reset your thumbnails, causing them to be recreated according to the global settings. I'm afraid I can't think of a way to prevent a specific directory from being thumbnailed.Note: In Ubuntu 12.04 (Precise Pangolin) and older, the thumbnails are stored in
~/.thumbnails. But please verify this for your own distro. Lubuntu 12.10 (a clean install) has its thumbnails in ~/.thumbnails and there is nothumbnailssubfolder in ~/.cache.
