Pressing the play button on earphones opens up iTunes. Is there a way I can prevent this from happening without installing any third party programs? I would preferably want to control Spotify with my earphones.

Yes, there is an option under Preferences -> Advanced Preferences, under the separator saying 'Keep movie window on top of all other windows'.
The window may look slightly different than this (I found this example online), but the wording is still the same.
You can do this with ControllerMate:
And a little little AppleScript (each line for each function):
tell application "Spotify" to playpause
tell application "Spotify" to next track
tell application "Spotify" to previous track
If you open QuickTime Player it will silently ignore the play/pause button and prevent iTunes from being opened, if you don't iTunes will still try to play something. Another solution to prevent iTunes from starting is to disable the 'Remote Control Daemon', how to do this is explained in detail on superuser.

Not if there's nothing currently playing. Play controls will try to resume the last thing you played that is still running and in memory. If the OS can't find anything to resume, it will default to iTunes.
