I would like to use the following 3 monitor setup because the arrangement aligns logically with the physical placement of the monitors.
Is it possible to bridge the gap between the left and right monitors, above the primary?
OR
Is it possible to detect and move the mouse via scripting?
You don't need to worry about Spaces for this. What you need to do is go into:
System Preferences --> Displays --> Arrangement
Drag the icon representing your external monitor to the top of the icon representing your macbook monitor:
You can also drag the bar representing your menubar/dock to whichever monitor you want to be your "main" display.
This is not possible, the multi monitor support on Lion is ridiculously lacking, especially given Apple jumping up and down about how amazing Thunderbolts (and the displays) are.
If you want to use more than one screen, you are pretty much unable to use FullScreen. (without making the secondary screen useless)

Just thinking outside the box a minute…
…wouldn't this achieve something close to what you want, without the need for any external solution.
It would give you a clean 'jump' between the external screens, balanced against a slight loss of accuracy in the transition between the top left & your main screen.
