When I'm watching a movie and I press the "play" button, iTunes 10 launches. How can I keep this from happening?

The simplest way is to un-tick the Automatically sync this... box* on the Summary Screen for the device in iTunes when the device is connected:
- This is per device.
Alternatively you can just remove the iTunesHelper service under your Login Items which will disable it irrelevant of the tick box state.
You can do this with ControllerMate:
And a little little AppleScript (each line for each function):
tell application "Spotify" to playpause
tell application "Spotify" to next track
tell application "Spotify" to previous track
If you open QuickTime Player it will silently ignore the play/pause button and prevent iTunes from being opened, if you don't iTunes will still try to play something. Another solution to prevent iTunes from starting is to disable the 'Remote Control Daemon', how to do this is explained in detail on superuser.

Just found this on Lifehacker. I think it's exactly what you're looking for. The post talks about a patch called MMFix, which changes the media control daemon to stop exactly this from happening. Apparently, it comes with a one-click installer.
And, if some reason you would like to remove the patch, just run the one-click installer again and the original rcd daemon will be restored.
(Works on OS X 10.7 Lion, too!)
